Comparison of HPLC methods in HbA1c determination |

Objective: Method comparison experiments are based on comparison of samples results analyzed by
the new test method and previously accepted method. In this study is aimed to compare reversedphase
cation exchange chromatography with boronate affinity chromatography method which is
currently used in HbA1c measurements in our laboratory.
Materials and Methods: One hundred and thirty one patient samples which were sent to biochemistry
laboratory for HbA1c measurement were included in this study. HbA1c measurements were made in
Adams HA-8180V (Arkray, KDK, Shiga, Japan- reversed-phase cation exchange chromatography) and
Premier Hb9210 (Trinity Biotech, Ireland-boronate affinity chromatography) HPLC instruments.Method
comparison studies were made according to National Committee for Clinical Laboratory Standarts
(NCCLS) EP-9 A2 protocol which was published by Clinical Laboratory Standarts Institute.
Results: Results of 131 patient samples were recorded to SPSS 21.0 for method comparison. The
correlation between two methods was statistically significant (r= 0.980, p< 0.05) and the regression
equation was found as (y= 0.27+0.95x).
Conclusion: HbA1c measurements with two different methods were determined to be
compatible,accurate and reliable. Besides,we thought that it will be appropiate to consider other factors
such as speed and cost when selecting the methods for routine use.
