Oxidant - Antioxidant Parameters and Their Relationship in Patients with Gastric Cancer

Objective: We aimed to determine the relationship between some oxidative and antioxidant parameters and cancer status. Therefore, the activity of serum PON1, an enzyme known as a lipid antioxidant, as well as serum level of TBARS, the end product of lipid peroxidation, and thiol, a member of antioxidant team, were determined for evaluating oxidative stress in patients with advanced gastric cancer.

Materials and Methods: Thirty-three patients with advanced gastric cancer (stage IV) were included in the study and compared with healthy controls (n=24). Serum PON1 activity, TBARS and thiol levels were measured by enzymatic spectrophotometric methods.

Results: The serum TBARS level was higher in the patient group (2.24±0.9 nmol/mL) than controls (1.34±0.3 nmol/mL; p<0.001). PON1 activity was found to be lower in the patient group (150.62± 69.2 U/L) than the control group (304.95±112.4 U/L; p<0.05). Serum thiol level was found to be lower in the gastric cancer group (257.91±59.7 mmol/L) than the control group (319.18±118.8 mmol/L; p<0.05). Serum PON1 activity was found to be positively correlated with thiol level (r=0.345, p<0.05) and negatively correlated with TBARS level (r=-0.280, p<0.05).

Conclusion: These results indicate that increased TBARS levels may be cause of the decreased PON activity and thiol levels, which act as a tumor promoter and a co-carcinogenic agent, in patients with gastric cancer. Since PON1 is also an antioxidant agent, effective antioxidant therapy to inhibit lipid peroxidation and to increase PON1 activity may be an alternative adjunct therapeutic option in gastric cancer treatment.
