The Effect of Cryopreservation on Sperm Malondialdehyde Levels and Motility

Objective: Sperm cryopreservation is the procedure of freezing sperms without loosing their liveliness in various oncological and/or immunological diseases and fertilization problems arousing from the treatments of these diseases. However, a decrease in sperm motiliy, cell injury and an increase in free oxygen radicals have been reported after this procedure. In our study, we investigated the probable mechanism of negative effect in motility after cryopreservation.

Material and Methods: In this prospective study, we examined sperm motility, malondialdehyde (MDA) levels and the effects of MDA levels, which is the last product of lipid peroxidation, on motility in samples of 30 testis cancer patients who donated sperms for cryopreservation before freezing and melted after one month.

Results: After cryopreservation, while the MDA levels increased (p<0.005) sperm motility decreased (p<0.0001). According to Spearman correlation analysis, there was a correlation between the increase in MDA levels and the decrease in the sperm motility (before cryopreservation p= 0.03, after cryopreservation p= 0.001).

Conclusion: We suggest that cryopreservation causes a decreasing effect in sperm motility by increasing lipid peroxidation.
